Influence of Escherichia coli hydrogenases on hydrogen fermentation from glycerol

چکیده
Since the actual role of Escherichia coli hydrogenases on fermentation from glycerol has not been clear, we evaluated the effect of inactivation of each E. coli hydrogenase on cell growth, hydrogen production, organic acids production, and ethanol production. Inactivation of hydrogenase 2 and hydrogenase 3 reduced cell growth, hydrogen and succinate production as well as glycerol utilization while acetate increased. Inactivation of hydrogenase 2 in minimal medium at pH 7.5 impaired hydrogen production, but no significant effect occurred at pH 6.5 or in complex medium. Inactivation of hydrogenase 3 impaired hydrogen production in minimal and rich medium, pH 6.5 and pH 7.5 accumulating formate in all conditions. Therefore during fermentation from glycerol, hydrogenase 3 is the main hydrogenase with hydrogen synthesis activity through the formate hydrogen lyase complex. Hydrogenase 2 seems mainly required for optimum glycerol metabolism rather than hydrogen synthesis. There were no significant impacts by inactivating hydrogenase 1 and hydrogenase 4. Dissecting the roles of E. coli hydrogenases in biohydrogen production
Escherichia coli can perform at least two modes of anaerobic hydrogen metabolism and expresses at least two types of hydrogenase activity. Respiratory hydrogen oxidation is catalysed by two ‘uptake’ hydrogenase isoenzymes, hydrogenases -1 and -2, and fermentative hydrogen production is catalysed by hydrogenase-3. Harnessing and enhancing the metabolic capability of Escherichia coli to perform a...

متن کاملBiosynthesis of Ethanol and Hydrogen by Glycerol Fermentation Using Escherichia coli
Production of high value products from glycerol via anaerobic fermentation is of utmost importance for the biodiesel industry. The microorganism Escherichia coli (E. coli) K12 was used for fermentation of glycerol. The effects of glycerol concentration and headspace conditions on the cell growth, ethanol and hydrogen production were investigated. A full factorial experimental design with 3 repl...
متن کاملAnaerobic growth of Escherichia coli on glycerol by importing genes of the dha regulon from Klebsiella pneumoniae.
The dha regulon of Klebsiella pneumoniae specifying fermentative dissimilation of glycerol was mobilized by the broad-host-range plasmid RP4:mini Mu and introduced conjugatively into Escherichia coli. The recipient E. coli was enabled to grow anaerobically on glycerol without added hydrogen acceptors, although its cell yield was less than that of K. pneumoniae. The reduced cell yield was probab...
متن کاملProperties of cell-free hydrogenases of Escherichia coli and Rhodospirillum rubrum.
The mechanism of conversion of substrate hydrogen to H2 in microbial fermentations is still obscure. One of the enzymes presumably involved in the final phase of H2 formation is hydrogenase, which catalyzes the reaction H2 =2H+ + 2e (Stephenson and Stickland, 1931). The present investigation was stimulated by our interest in the presumed participation of hydrogenase in photochemical production ...
